Detection of BCAA in Biological Samples

Branched chain amino acids (BCAA; leucine, valine and isoleucine) have important roles in many areas of biology as substrates for protein synthesis, an energy source, and essential signaling molecules. The BCAA-Glo™ Assay is a rapid, sensitive method for BCAA detection based on bioluminescent technology, allowing you to detect the smallest changes of BCAAs in a variety of biological sample types.

How the BCAA-Glo™ Assay Works

The BCAA-Glo™ Assay uses leucine dehydrogenase to produce NADH in the presence of BCAA, followed by NADH measurement using a bioluminescent NADH detection technology. The result is a light signal proportional to the starting BCAA concentration in the sample.

Detecting BCAA using BCAA-Glo™ Assay is simple—just add Detection Reagent to samples and incubate. The luminescent output can be analyzed using a standard plate-reading luminometer. The assay is compatible with cell lysates, media samples, tissue homogenates, plasma and serum samples.

Leucine titration curve. The bioluminescent signal (RLU) is proportional to BCAA content of the sample. Precise concentration of BCAA in a given sample can be determined by running a standard curve with the leucine positive control provided in the kit.

In-Well Detection is Easy and Sensitive

For intracellular BCAA measurements, sample preparation steps are streamlined to enable cell lysis and subsequent BCAA detection in the same well. No cell collection, centrifugation or spin columns are needed. Simply remove medium from plated cells and wash with PBS, and then add acid to lyse cells. The lysate can be assayed directly in the well or after being transferred to a new 96- or 384-well plate.

In-well detection of BCAAs using the BCAA-Glo™ Assay.

A small number of cells is required for in-well BCAA detection. A549 cells were dispensed into wells of a 96-well plate in the indicated quantities in quadruplicate. Following in-well lysis, BCAA content in each sample was measured with BCAA-Glo™ Assay.
