Quantify Inflammasome Activation Without Tedious Wash Steps

The Lumit™ IL-1β Immunoassay quantitatively measures released IL-1β in cell culture samples using a simple, no-wash protocol. Just add labeled antibodies to the sample, add detection reagent and read luminescent signal using a standard plate-reading luminometer. The entire protocol is completed in less than 70 minutes! The assay can be used directly on cells in culture or culture medium transferred to a separate assay plate.

How the Lumit™ IL-1β Immunoassay Works

Primary antibodies to IL-1β were selected for their specific and sensitive detection and labeled with the LgBiT and SmBiT subunits of NanoBiT® Luciferase. In the presence of IL-1β, the subunits are brought together to form an active luciferase enzyme. Addition of optimized substrate generates a bright luminescent signal proportional to IL-1β levels.

High-Throughput NLRP3 Inflammasome Inhibitor Screening

The add-and-read format of the Lumit™ IL-1β Immunoassay enables high-throughput screening for modulators of IL-1β release.
IL-1b inhibitor high throughput screening

The NLRP3 inflammasome inhibitor, MCC950, prevents caspase-1 activation and release of mature IL-1β. A titration of MCC950 was added to differentiated THP-1 cells (5 × 104/well) followed by LPS (20 EU/ml) or vehicle. The Lumit™ IL-1β (Human) Immunoassay was performed after 5 hours.
