Homogeneous Assay Measures Caspase-3/7 Activity

The Caspase-Glo® 3/7 assay provides a proluminescent caspase-3/7 DEVD-aminoluciferin substrate and a proprietary thermostable luciferase in a reagent optimized for caspase-3/7 activity, luciferase activity and cell lysis. Adding the single Caspase-Glo® 3/7 Reagent in an "add-mix-measure" format results in cell lysis, followed by caspase cleavage of the substrate. This liberates free aminoluciferin, which is consumed by the luciferase, generating a "glow-type" luminescent signal that is proportional to caspase-3/7 activity.

The stabilized luciferase and proprietary buffer system improve assay performance across a wide range of assay conditions, and the assay is less likely to be affected by compound interference than fluorescent- or colorimetric-based assays. The caspase activity assay is designed for use with multiwell plate formats using either purified enzyme or cells in culture and can be multiplexed with other cell-based assays. High sensitivity allows use of the Caspase-Glo® 3/7 assay in 3D cell culture systems. The assay delivers excellent Z'-factor values in cell and purified enzyme models.

Multiplex Caspase 3/7 and Cytotoxicity Assays in 3D Cultures

HCT116 spheroids were created using the the InSphero GravityPLUSTM Hanging Drop System, and then treated with Panobinostat for up to 72 hours. CellToxTM Green was added at time of treatment. At 72 hours Caspase-Glo® 3/7 Reagent was added and plates were shaken for 30 minutes. Fluorescence was read at 485nm excitation, 520nm emission, followed by luminescence.

Using 100μl of Caspase-Glo® Reagent per assay in a 96-well format, Cat.# G8090 provides sufficient reagents to perform 25 caspase activity assays; Cat.# G8091, 100 assays; Cat.# G8092 and G8093, 1,000 assays. Using 25μl of Caspase-Glo® Reagent per assay in a 384-well format, Cat.# G8090 provides sufficient reagents to perform 100 caspase activity assays; Cat.# G8091, 400 assays; Cat.# G8092 and G8093, 4,000 assays.
