Detecting Drug:Target Interactions with BRET

The development of molecules that can bind to and modulate the activity of a target protein is critical for understanding the biology and treatment of disease. Using Bioluminescence Resonance Energy Transfer (BRET), it is possible to quantitatively measure the interaction between a molecule and a protein in live cells. In some cases it is even possible to measure these interactions in real-time, observing not only the formation of the interaction but also the dissociation of the partners.

The study of Ligand:Protein Interactions with BRET relies on the energy from a luciferase tagged protein to act as a BRET donor, and this energy is transferred to a fluorescently tagged ligand acting as a BRET acceptor. This energy transfer is moderated by the proximity of the two partners. NanoLuc® Luciferase offers unique advantages as a BRET donor due to its very bright light output and small size. NanoLuc® Luciferase can use a variety of fluorophores as BRET acceptors.
