Measure Compound Affinity and Occupancy at Multimeric RAS in Live Cells

The NanoBRET® TE Intracellular RAS Assay is a novel method for measuring intracellular compound binding to the RAS proteins, a family of GTPases that are frequently mutated in human cancers. This assay is based on NanoBRET® TE technology, an energy transfer technique designed to measure molecular proximity in living cells, and can be used to quantify compound affinity and fractional occupancy for multiple RAS variants, including KRAS, HRAS and related mutants. Instead of using NanoLuc® luciferase-fused target protein as the BRET donor, the NanoBRET® TE Intracellular RAS Assay uses the NanoLuc® Binary Technology (NanoBiT) to enable the measurement of cellular target engagement at multimeric RAS.

How the NanoBRET® TE RAS Assay Works

The NanoBRET® TE Intracellular RAS Assay uses NanoBiT® Technology, a structural complementation system comprised of a large luciferase subunit (LgBiT) and a small complementary peptide (SmBiT(q)). Specifically, a fusion protein of LgBiT and full-length RAS is co-expressed with a fusion protein of SmBiT(q) and full-length RAS in cells. When the RAS proteins interact and form multimers, the LgBiT and SmBiT(q) subunits come together to form an active luciferase enzyme that serves as the BRET donor. The energy acceptor is a cell-permeable fluorescent NanoBRET® tracer that binds specifically to RAS. Addition of compounds that bind to RAS displaces the tracer and results in a dose-dependent loss of BRET signal.

The NanoBiT® system used in the NanoBRET® TE Intracellular RAS Assay is unique in the use of the SmBiT(q) peptide, which makes the NanoBiT® luciferase enzyme sensitive to the Extracellular NanoLuc® Inhibitor. This inhibitor quenches signal that can arise from compromised cells and cellular debris to ensure the NanoBRET® signal originates from live cells.

Measure Orthosteric and Allosteric RAS Ligands Across Multiple RAS Variants

The NanoBRET® TE Intracellular RAS Assay can be used to measure compound cellular affinity and occupancy for multiple RAS proteins, including wildtype and mutant KRAS/HRAS. The assay can detect compound binding to both switch I/II pocket and switch II pocket.

The switch-II pocket (SII-P) is targeted by the current small-molecule inhibitors of KRAS(G12C), which exploit the nucleophilicity of the cysteine mutation and engage the inactive GDP state of KRAS(G12C). However, not all oncogenic KRAS mutants share these two features, and it remains unknown whether SII-P is a viable target for KRAS mutants beyond G12C. In this study published in Nature Chemical Biology, NanoBRET® Target Engagement Assays were used in combination with NMR spectroscopy to interrogate the accessibility of SII-Ps for non-G12C mutants of KRAS. The results show broad opportunity for targeting SII-P across many KRAS hotspot mutants.
