Homogeneous Assay Measures Caspase-3/7 Activity
The Caspase-Glo® 3/7 Assay provides a proluminescent caspase-3/7 DEVD-aminoluciferin substrate and a proprietary thermostable luciferase in a reagent optimized for caspase-3/7 activity, luciferase activity and cell lysis. Adding the single Caspase-Glo® 3/7 Reagent in an "add-mix-measure" format results in cell lysis, followed by caspase cleavage of the substrate. This liberates free aminoluciferin, which is consumed by the luciferase, generating a "glow-type" luminescent signal that is proportional to caspase-3/7 activity.
The stabilized luciferase and proprietary buffer system improve assay performance across a wide range of assay conditions, and the assay is less likely to be affected by compound interference than fluorescent- or colorimetric-based assays. The caspase activity assay is designed for use with multiwell plate formats using either purified enzyme or cells in culture and can be multiplexed with other cell-based assays. The assay delivers excellent Z´-factor values in cell and purified enzyme models. Using the MyGlo™ Reagent Reader in conjunction with the ProNect™ Data Platform, you can visualize caspase activity with ease.

Simple Bioluminescent Detection of Caspase Activity
Luminescence Is Linear Over a Broad Range of Cell Numbers
Jurkat cells were treated with anti-Fas mAb for 4.5 hours to induce apoptosis or were left untreated. Caspase-Glo® 3/7 Reagent was added directly to the cells in 96-well plates and incubated for 1 hour before recording luminescence. Each point represents the average of 4 wells. The "no cell" blank control value has been subtracted from each.
Measure Dose Response to Apoptosis Inducers
Jurkat cells were seeded at 10,000 cells/well and treated for 20 hours with two compounds. Caspase-Glo® 3/7 Reagent was added to the cells and luminescence measured on the MyGlo™ Reagent Reader. The dose-response graph shows an apoptotic response to bortezomib with no apoptotic response to palbociclib.
