Lumit™ Technology: Reliable, Automation-Friendly Immunodetection

Lumit™ Technology enables bioluminescence-based detection of specific antigens or antibodies, simplifying immunoassay protocols and speeding time to results. Based on the award-winning NanoBiT® complementation system, Lumit™ Technology powers sensitive and accurate immunoassays for research and clinical applications.

Lumit™ Immunoassays provide a faster, more reliable alternative to ELISAs, eliminating the variability associated with multiple wash steps, and providing the convenience of a simple and sensitive luminescent detection method. Because of these advantages, Lumit™ Immunoassays are easily adaptable to automation for labs processing large numbers of samples.

NanoBiT® Technology is the Basis of the Lumit™ Detection Method

NanoLuc® Binary Technology (NanoBiT) is a protein complementation system based on NanoLuc® luciferase, which consists of two subunits—small bit (SmBiT) and large bit (LgBiT)1,2. These subunits have low affinity for each other and only form an active luciferase enzyme when they come into close proximity. The small size of the NanoBiT® complementation partners minimizes interference with normal protein function, and the bright signal accommodates low, native expression levels.

Antigen Detection

In some Lumit™ Immunoassays, antibodies directed against specific target molecules are chemically labeled with SmBiT and LgBiT. When the two antibodies bind their target, SmBiT and LgBiT come into close proximity to form an active luciferase, generating a luminescence signal in the presence of substrate3

Antibody Detection

Lumit™ Technology can also be used to detect antibodies. The Lumit™ SARS-CoV-2 Immunoassay uses a fragment of the SARS-CoV-2 spike protein receptor binding domain (RBD) labeled with SmBiT and LgBiT. When the labeled viral protein fragments are incubated with a sample containing SARS-CoV-2 antibodies, they bind to the antibody, bringing the SmBiT and LgBiT subunits together to generate a luminescence signal.

The Lumit™ Dx SARS-CoV-2 Immunoassay is available as a diagnostic assay in the United States. The assay is also ideal for research applications such as population surveillance, epidemiological studies and vaccine research.
