The mFcγRIV ADCC Reporter Bioassay, Core Kit, is a bioluminescent cell-based assay that can be used to measure the potency and stability of antibodies and other biologics with Fc domains that specifically bind and activate murine FcγRIV (mFcγRIV).  mFcγRIV is the predominant receptor involved in ADCC in the mouse and is more closely related to human FcγRIIIa, the primary Fc receptor involved in ADCC in humans, than mFcγRIII. The assay consists of a genetically engineered Jurkat T cell line that expresses: 

  • Mouse FcγRIV receptor
  • A luciferase reporter driven by an NFAT-response element (NFAT-RE)

The mFcγRIV Effector Cells are provided in thaw-and-use format, which includes cryopreserved cells that can be thawed, plated and used in an assay without the need for propagation. When co-cultured with a target disease cell and relevant antibody, the mFcγRIV Effector Cells bind the Fc domain of the antibody, resulting in mFcγRIV signaling and NFAT-RE-mediated luciferase activity. The bioluminescent signal is detected and quantified using Bio-Glo™ Luciferase Assay System (Cat.# G7940, G7941) and a standard luminometer such as the GloMax® Discover System.
