Notes: The Wizard® Plus Minipreps DNA Purification Resin was used to isolate apoptotic DNA from primary cultures of cerebellar granule cells. The cells were lysed with 7M Guanidine HCl, mixed with 1ml of resin, spun down, resuspended in wash solution, collected in a minicolumn, washed and eluted in water. The DNA was treated with RNase A and treated with the nucleic acid stain, SYBR Green, for quantitation versus a DNA standard. Two hundred nanograms of the isolated DNA were analyzed by agarose gel electrophoresis and ethidium bromide staining. The RQ1 RNase-Free DNase was used to treated total RNA samples prior to RT-PCR.
