State-of-the-Art Dual-Reporter Technology brings Greatest Sensitivity and Flexibility

The Nano-Glo® Dual-Luciferase® Reporter (NanoDLR™) Assay detects the activities of dual luciferases, firefly and NanoLuc® reporter enzymes, in a single sample. Improved firefly chemistry and the bright, sensitive NanoLuc® luciferase make this a highly sensitive dual-luciferase assay that is simple to use, provides superior data quality, and allows greater flexibility in assay design.

In the NanoDLR™ Assay protocol, firefly luciferase activity is measured first using the ONE-Glo™ EX Luciferase Assay Reagent. NanoDLR™ Stop & Glo® Reagent is then added to quench the firefly signal and provide the substrate needed to measure NanoLuc® luciferase activity.

Bright, Stable Signal

When dual luciferases are expressed from the same promoter, the NanoDLR™ Assay provides bright signal for both Fluc and Nluc reporters with stable "glow-type" signals. The signal half-life is greater than two hours for both reporters.

Here, we compared luminescence signals from HEK293 cells transfected with a 1:1:8 ratio of either TK-Rluc (Renilla):TK-Fluc (firefly):carrier DNA or TK-Nluc (NanoLuc):TK-Fluc:carrier DNA and assayed with NanoDLR™, DLR™ or Dual-Glo® Dual-Luciferase Assay Systems as indicated.  

Better Quenching, Greater Sensitivity

Potent inhibition of firefly luciferase (Fluc) provides excellent signal separation, allowing accurate measurement of even low amounts of NanoLuc® luciferase (Nluc), even in the presence of high Fluc activity.

This data shows the efficient quenching of the firefly luciferase signal in the NanoDLR™ Assay. Even extremely high Fluc signals are reduced to near background levels by NanoDLR™ Stop & Glo® Reagent, providing excellent signal separation and low variability without sacrificing the sensitivity of two bright luminescent readouts.

Dual-luciferase assay with firefly reporter and NanoLuc control

Two Primary Reporters

Measure two different biological responses simultaneously.

  • Best for: Measuring 2 pathways at once for maximum data; multiplexing protein stability and genetic reporter assays.
  • Reporter Constructs: NlucP Experimental Reporter; FlucP Experimental Reporter; Nluc protein stability reporter.

In this example, the NanoDLR™ Assay was used to measure protein dynamics and gene activation in the same sample. HEK293 cells were transiently transfected with the pNLF1-HIF1A[CMV/neo] fusion construct, diluted 1:1000 into the Hypoxia Response Element Vector, pGL4.42[luc2P/HRE/Hygro]. After 18 hours, cells were stimulated with varying doses of 1,10-phenanthroline and assayed for expression of the firefly luciferase transcriptional reporter and the HIF1a-NanoLuc® fusion protein using the Nano-Glo® Dual-Luciferase® Reporter (NanoDLR™) Assay at the indicated time points.

Improved Reagent Stability Simplifies Repeat Use and Reduces Waste

The reconstituted ONE-Glo™ EX Reagent in the NanoDLR™ Assay has increased stability at room temperature and 7°C , reducing the need to dispense into aliquots and freeze reagent between uses.

The data to the right illustrate NanoDLR™ Reagents extended stability once reconstituted. Reconstituted reagents were stored at either 22°C or 7°C and frozen at –70°C at defined times. Upon thawing, reagents were combined with purified firefly or NanoLuc® luciferase and the relative activity calculated as the luminescence signal intensity for each sample measured 3 minutes after enzyme addition, relative to the signal intensity of the sample that was placed at –70°C with no 22°C incubation; n=3.

Researchers shall have no right to modify or otherwise create variations of the nucleotide sequence of the luciferase gene except that researcher may (1) create fused gene sequences provided that the coding sequence of the resulting luciferase gene has no more than four deoxynucleotides missing at the affected terminus compared to the intact luciferase gene sequence; and (2) insert and remove nucleic acid sequences in splicing research predicated on the inactivation or reconstitution of the luminescence of the encoded luciferase. No other use of this product or derivatives is authorized without the prior express written consent of Promega.

In addition, researchers must:
(1a) use Nano-Glo®-branded luminescent assay reagents (LARs) for all determinations of luminescence activity of this product and its derivatives; or
(1b) contact Promega to obtain a license for use of the product and its derivatives with LARs not manufactured by Promega.

For uses of Nano-Glo®-branded LARs intended for energy transfer (such as bioluminescence resonance energy transfer) to acceptors other than a genetically encoded autofluorescent protein, researchers must:
(2a) use NanoBRET™-branded energy acceptors (e.g., BRET-optimized HaloTag® ligands) for all determinations of energy transfer activity by this product and its derivatives; or
(2b) contact Promega to obtain a license for use of the product and its derivatives for energy transfer assays to energy acceptors not manufactured by Promega.

Researchers may transfer derivatives to others for research use provided that at the time of transfer a copy of this label license is given to the recipients and recipients agree to be bound by the terms of this label license. With respect to any uses outside this label license, including any diagnostic, therapeutic, prophylactic or commercial uses, please contact Promega for supply and licensing information.
