Overview

Lumit® Second Messenger Assays offer robust, no-wash luminescent competitive binding methods ideal for quantifying intracellular cyclic nucleotides (cAMP, cGMP or cGAMP) directly from cell lysates. Utilizing Promega's innovative Lumit® technology based on NanoLuc® Luciferase structural complementation technology, these assays deliver sensitive detection with streamlined workflows suitable for high-throughput screening and drug discovery applications.

Assay Workflow

The Lumit® cyclic nucleotide assays include a tracer composed of cyclic nucleotide labeled with SmBiT (Tracer), a primary mouse anti-cyclic nucleotide monoclonal antibody, and a secondary anti-mouse antibody conjugated to LgBiT. In the absence of unlabeled cyclic nucleotide, the tracer binds the antibodies to form a complex, bringing SmBiT and LgBiT into close proximity and reconstituting the Nanoluc® luciferase. Upon addition of Lumit® Detection Substrate, this interaction generates maximal luminescent signal output. When unlabeled cyclic nucleotide is present in increasing concentrations, it competes with the tracer for antibody binding, resulting in a reduction of luminescent signal proportional to the unlabeled cAMP concentration.
