HaloTag® technology is a comprehensive platform for functional protein analysis based on the formation of a covalent bond between a protein fusion tag (HaloTag®) and synthetic chemical ligands. By using different ligands you can control the function and properties of the HaloTag® fusion protein. The HaloTag® system gives exceptional performance in cell-based and biochemical assays without the need to switch fusion tags or reclone inserts.

These custom assay materials are developed by our Custom Assay Services team within R&D. Although functionally tested, they are not manufactured under the Promega ISO13485 quality system and are sold “as is” with no warranty either stated or implied.

Custom HaloTag® Expression Vectors

N-terminal and C-terminal Flexi® HaloTag® Vectors for attenuated expression, protein secretion, and nuclear localization in mammalian cells, and vectors containing regulated mammalian, TK, SV40, PGK, EF1α promoters are available.

Custom HaloTag® Ligands

  • pH sensor ligand to detect internalization of membrane proteins
  • Eosin ligand for chromophore-assisted laser inactivation (CALI) for targeted protein destruction
  • HaloTag PROTAC (proteolysis targeting chimera) is a permeable small molecule compound which can target HaloTag fusion proteins for degradation. HaloTag PROTAC will bind covalently to HaloTag and induce interactions with E3 ligase machinery proteins, resulting in ubiquitination of the HaloTag fusion and subsequent trafficking to the proteasome for degradation.
