ViaScript® (HiBiT) TCK Bioassay: Screen. Select. Success.

We know how critical and frustrating it can be for pharmaceutical and biotech companies to rigorously screen cell lines and clones for target expression to ensure validity, data reproducibility and regulatory compliance. If you pick the wrong cell line, it can lead to costly experimental failures and misleading data.

However, you don’t need to spend weeks selecting a stable clone that meets the necessary criteria to move forward, which can be time-consuming, labor intensive and constrained by the variability and limitations of traditional assays. Principle of ViaScript® (HiBiT) TCK Bioassay. The ViaScript® Reagent allows researchers to transiently transfect HiBiT mRNA into a cell line of interest to enable early screen and clone selection. The HiBiT cell line can then be used in a HiBiT TCK Bioassay and paired with qualified primary effector cells and a biologic to measure ADCC, ADCP, TDCC or CAR-T cell killing. Killing of the target cell releases HiBiT protein into the media, and substrate addition allows HiBiT to bind with high affinity to its LgBiT counterpart to form the functional NanoBiT® luciferase. The resulting luminescent signal is proportional to the number of dead cells in the culture and is specific to target cell death.
