Principle of Bioassays for Incretin-Related Biologics
- The GLP-1, GIP and Glucagon Bioassays use engineered cells to link hormone receptor activation to a luminescent reporter, providing a sensitive and quantitative readout of receptor activity.
- When incretin-related hormones bind to their receptors, they trigger receptor activation and intracellular signaling pathways through adenylyl cyclase, which produces cAMP and induces transcriptional activation of a response element (RE), leading to luciferase expression and bioluminescent signal.

Principle of Myostatin Bioassay
Myostatin is of significant interest to scientists developing obesity therapeutics due to its natural role as a negative regulator of muscle growth and differentiation. By inhibiting myostatin, researchers aim to increase muscle mass, which can enhance metabolic activity, improve insulin sensitivity and elevate basal metabolic rate—factors crucial for combating obesity. Representation of the Myostatin Bioassay
The Myostatin Bioassay consists of a genetically engineered cell line, Myostatin Bioassay cells. When ActRIIA/B ligands bind to the receptor, they trigger receptor activation and intracellular signaling pathways through SMAD signaling pathways, which triggers transcriptional activation of a response element (RE), leading to NanoLuc® Luciferase expression and bioluminescent signal.
Myostatin Bioassay: Performance Data
The Myostatin Bioassay is suitable for demonstrating activity of ligands and antibodies directed toward myostatin receptor binding. Panel A. Myostatin Bioassay cells were treated with titrations of activin or myostatin. Panel B. Myostatin Bioassay cells were treated with titrations of anti-myostatin blocking antibody, landogrozumab, in the presence of EC80 of myostatin. In both panels, after 5 hours, Bio-Glo-NL™ Reagent was added and plates were read on a GloMax® Discover Microplate Reader.

5. For uses of this material for energy transfer (such as bioluminescence resonance energy transfer), recipient must:
- 5.1. Use NanoBRET™-branded luminescent assay reagents (LARs; e.g., NanoBRET™ Nano-Glo® Substrate), Intracellular TE Nano-Glo® Substrate/Inhibitor or Intracellular TE Nano-Glo® Vivazine™/Inhibitor for all determinations of luminescent activity by this material and its derivatives; and
- 5.2. Use NanoBRET™-branded energy acceptors (e.g., NanoBRET™ tracers, NanoBRET™ dyes, BRET-optimized HaloTag® ligands) for all determinations of energy transfer activity by this material and its derivatives; or
- 5.3. Contact Promega to obtain a license for the use of the material and its derivatives for energy transfer assays to energy acceptors not manufactured by Promega. No license is needed if the energy transfer acceptor is a genetically encoded autofluorescent protein.

6. For uses of HaloTag® Technology in this material, recipient must either:
- 6.1. Use Promega HaloTag® ligands, which can be modified or linked to Promega or customer-supplied moieties; or
- 6.2. Contact Promega to obtain a license if Promega HaloTag® ligands are not to be used.
