Croat. Med. J. 46, 563–577. Improving efficiency of a small forensic DNA laboratory: validation of robotic assays and evaluation of microcapillary array device. 2005

Crouse, C.A., Yeung, S., Greenspoon, S., McGuckian, A., Sikorsky, J., Ban, J. and Mathies, R.

Notes: The authors validated the DNA IQ™ System for manual and automated DNA purification from blood, tissue, bone, hair, epithelial cells and mixed stains such as semen. Automated DNA extraction was performed using the Beckman Coulter Biomek® 2000 workstation. Purified DNA (0.5–1.0ng) was then amplified using the PowerPlex® 16 BIO System and a 6% PAGE PLUS™ gel. Automated DNA purification and use of a single-amplification STR system greatly increased sample throughput. (3645)

J. Forensic Sci. 49, 71–80. Validation and implementation of the PowerPlex™ 16 BIO System STR multiplex for forensic casework. 2004

Greenspoon, S.A., Ban, J.D., Pablo, L., Crouse, C.A., Kist, F.G., Tomsey, C.S., Glessner, A.L., Mihalacki, L.R., Long, T.M., Heidebrecht, B.J., Braunstein, C.A., Freeman, D.A., Soberalski, C., Bruesehoff, N., Amin, A.S., Elizabeth K. Douglas E.K. and Schumm, J.W.

Notes: A concordance study was performed by several independent agencies using the PowerPlex® 16 BIO System. The study tested the ability of the PowerPlex® 16 BIO System to type 653 samples previously typed using the PowerPlex® 1.1 and 2.1 Systems. The authors discuss stutter, sensitivity and precision. Studies were also performed with mixed samples, nonhuman DNA samples, and samples exposed to various environmental conditions. (3008)

J. Forensic Sci. 47, 773-785. Validation of a 16-locus fluorescent multiplex system. 2003

Krenke, B.E., Tereba, A., Anderson, S.J., Buel, E., Culhane, S., Finis, C.J., Tomsey, C.S., Zachetti, J.M., Masibay, A., Rabbach, D.R., Amiott, E.A. and Sprecher, C.J.

Notes: In this paper, researchers from both Promega and state crime laboratories provide an evaluation of the Powerplex® 16 System loci (CODIS, Penta D, Penta E, and amelogenin). Data from several laboratories using a variety of thermal cyclers and the ABI PRISM® 310 Genetic Analyzer or ABI PRISM® 377 DNA Sequencer are presented. The Powerplex® 16 System was able to consistently genotype samples from as little as 0.0625-2ng of genomic DNA template. Other factors such as reaction volume, annealing temperature, titration of AmpliTaq Gold® DNA Polymerase, magnesium, or primer pairs, and 1ng DNA mixtures were analyzed using the Powerplex® 16 System.  The researchers also present data on stutter analysis and cross reactivity of primer sets on primate DNA templates for each loci. J. Forensic Sci. 46(4), 998-999. Allele frequencies for the PowerPlex® 16 STR loci in an Albanian population sample from northern Italy. 2001

Robino, C., Gino, S. and Torre, C.

Notes: The authors performed a population study using the PowerPlex® 16 System. (2277)

J. Forensic Sci. 46(3), 637-641. Concordance study on population database samples using the PowerPlex™ 16 kit and AmpFLSTR® Profiler Plus™ kit and AmpFLSTR® COfiler™ kit. 2001

Budowle, B., and Sprecher, C.J.

Notes: The PowerPlex® 16 System was used in a concordance study involving more than 500 population database samples that included African Americans, Bahamians, and Southwestern Hispanics. Samples were typed using PowerPlex® 16 and the Profiler Plus™ and COfiler™ kits. The results show that the primers used in these systems are reliable for typing reference samples destined for use in CODIS. Notes: This study reports that the primers used in the PowerPlex® 16 System are reliable for typing reference samples that are used in the FBI CODIS database. The authors also report allele frequency data for the Penta D and Penta E loci. (2275)

Moretti, T.R., Baumstark, A.L., Defenbaugh, D.A., Keys, K.M., Smerick, J.B. and Budowle B.

Notes: In this study, the amplification and typing conditions for the 13 core CODIS loci and their forensic applicability were evaluated. These loci are CSF1PO, FGA, TH01, TPOX, vWA, D3S1358, D5S818, D7S820, D8S1179, D13S317, D16S539, D18S51, and D21S11. The PowerPlex® Systems were evaluated, along with other commercially available STR typing systems. (2267)
