Fast Process and Flexible Protocols

The PowerPlex® 21 System is a multiplex STR system for human identification applications including forensic analysis, relationship testing and research use. The system allows co-amplification and fluorescent detection of 21 loci (20 STR loci and Amelogenin), including D1S1656, D2S1338, D3S1358, D5S818, D6S1043, D7S820, D8S1179, D12S391, D13S317, D16S539, D18S51, D19S433, D21S11, Amelogenin, CSF1PO, FGA, Penta D, Penta E, TH01, TPOX and vWA. The system offers high discrimination with 21 markers for difficult cases and complete data overlap with most existing multiplexes.

The PowerPlex® 21 System increases laboratory productivity and decreases average turnaround time with 90-minute PCR. Laboratories can also save labor and time since the wash step for FTA® cards is eliminated. Simpler protocols are available for swabs and nonFTA card punches as well. The higher inhibitor tolerance of this system benefits challenging casework samples and results in less locus drop-out and reaction failure for higher success rates.

10247TA_900px
A single-source template DNA (0.5ng) was amplified using the PowerPlex® 21 System and analyzed with an Applied Biosystems 3130 Genetic Analyzer using a 3kV, 5-second injection. Results were analyzed using GeneMapper® ID software, version 3.2.
ForensicGrade

Promega forensic products are manufactured in alignment with the ISO 18385 standard. This standard ensures minimal risk of human DNA contamination for products used to collect, store and analyze biological materials for forensic purposes. Learn more.

Flexibility for Most Sample Types

The PowerPlex® 21 System is designed to work with a variety of sample types, including difficult casework samples and is compatible with direct amplification from FTA® card punches as well as pretreated swabs and Bode Buccal Collector™ device or S&S 903 punches.

Genomic DNA
10363TA_900px
Amplification of 500pg of human genomic DNA using the recommended protocol. Amplified products were separated on an Applied Biosystems 3130 Genetic Analyzer (3kV, 5-second injection).
Blood on FTA® Card
10364TA_900px
Direct amplification of one 1.2mm punch from blood on an FTA® card using recommended protocol. Amplified products were separated on Applied Biosystems 3130 Genetic Analyzer (3kV, 5-second injection).
Buccal Samples on S&S 903
10365TA_900px
A 1.2mm punch from buccal samples on S&S 903 was pre-treated with SwabSolution™ Reagent, then amplified using the recommended protocol. Amplified products were separated on an Applied Biosystems 3130xl Genetic Analyzer (3kV, 5-second injection).

Swab from Buccal Samples

10370TA-W
A swab from buccal samples was pre-treated with SwabSolution™ Reagent, then amplified using the recommended protocol. Amplified products were separated on an Applied Biosystems 3130xl Genetic Analyzer (3kV, 5-second injection).

Rapid Processing Solutions for PowerPlex®

The 5X AmpSolution™ Reagent, SwabSolution™ Kit and PunchSolution™ Kit allow fast and simple processing of swabs and punches for analysis with various PowerPlex® Systems, including the PowerPlex® Fusion, 21, 18D, 16 HS, ESX, ESI, CS7 and Y23 Systems. These products are intended for preparation of single-source reference, database and paternity samples where sample purification is unnecessary.
