Are Promega lysis buffers compatible with Western blotting?

Yes, we have found four lysis buffers (Cell Culture Lysis Reagent [CCLR], Reporter Lysis Buffer [RLB], Passive Lysis Buffer [PLB] and Glo Lysis Buffer [GLB]) to be compatible with Western blotting. Antibody staining for cytoplasmic and nuclear proteins was successful with all lysis buffers although a freeze/thaw cycle enhanced cell lysis in some cases. However, only lysates made using GLB or CCLR were successfully probed for mitochondrial proteins. The experiment was performed using HeLa cells with 1X, 2X or 5X concentrations of each lysis buffer. Results may vary with other buffer concentrations and cell lines.
