How much protein will be synthesized in a TNT® Coupled Reticulocyte Lysate System reaction?

Yields will vary considerably, depending upon the protein being produced and the DNA template. For example, the presence of a poly(A) tail, sequences in the 5´ and 3´ untranslated regions, the context of the ATG and the presence of additional ATG codons upstream of the true translation initiation site can affect protein yield. As a general guideline, the positive control plasmid supplied with the kit generates 150–300ng of luciferase protein in a standard reaction.
