Functional Detection of ssRNA and Small-Molecule Agonists
TLR7 and TLR8 are endosomal pattern-recognition receptors that recognize single-stranded RNA (ssRNA) and small-molecule agonists, playing a key role in antiviral immunity and innate immune activation. Bioassays targeting TLR7 and TLR8 provide mechanism-of-action-based platforms for the detection and quantitation of agonist activity in research, drug development and quality control settings. These assays enable precise functional readouts of receptor activation, offering biologically relevant data that cannot be captured by physicochemical methods alone.
Assay Utility and Format Advantages
Available in thaw-and-use reporter cell formats, TLR7 and TLR8 bioassays streamline immunostimulatory profiling by eliminating the need for primary cells or complex culture systems. Their sensitivity to ssRNA structure, sequence context and delivery formulations makes them ideal for screening adjuvants, characterizing innate immune modulators and assessing off-target immunogenicity in RNA-based therapeutics. These bioassays facilitate robust, reproducible assessments of TLR-mediated activity, supporting both early-stage discovery and lot-release testing.
Single-Stranded mRNA Detection and Functional Activity
Principle of TLR7 and TLR8 Bioassays. TLR7 and TLR8 reporter cells uptake ssRNA or other synthetic TLR7 and TLR8 agonist, respectively, through endocytosis. The binding of the ssRNA to endosomal TLR7 or TLR8 leads to the activation of the TLR7- or TLR8-responsive promoter, which drives luciferase reporter expression and luminescent output.

Assays are Specific to TLR7 or TLR8 Activation
The TLR7 Bioassays and TLR8 Bioassays are specific to ssRNA detection. The TLR7 Bioassay effector cells (Panel A) and TLR8 Bioassay effector cells (Panel B) were incubated with increasing concentrations of the agonist R848. Luminescence was measured as a reporter of activation with parental cells lacking TLR7 or TLR8 serving as negative controls. Specific signal was observed only in receptor-expressing effector cells, confirming assay specificity to TLR7 or TLR8 activation.
Assays are Sequence- and Size-Specific
TLR7 and TLR8 Bioassays detect ssRNA in a sequence- and size-dependent manner. ssRNAs of various length were complexed with lipid transfection reagents and tested in the TLR7 (Panel A) and TLR8 (Panel B) Bioassays. ssRNA sequence length from longest to shortest is as follows: Firefly Luciferase > Poly(U) > ssRNA40 > ORN 06. Size-dependent differences were seen with TLR7 responding to the longer and more “U”-rich sequences. ssRNAs were either vendor-formulated or complexed in-house with Lipofectamine™ MessengerMAX™. Data demonstrate that TLR7 and TLR8 activation is influenced by both sequence and length of the RNA stimulus.

5. For uses of this material for energy transfer (such as bioluminescence resonance energy transfer), recipient must:
- 5.1. Use NanoBRET™-branded luminescent assay reagents (LARs; e.g., NanoBRET™ Nano-Glo® Substrate), Intracellular TE Nano-Glo® Substrate/Inhibitor or Intracellular TE Nano-Glo® Vivazine™/Inhibitor for all determinations of luminescent activity by this material and its derivatives; and
- 5.2. Use NanoBRET™-branded energy acceptors (e.g., NanoBRET™ tracers, NanoBRET™ dyes, BRET-optimized HaloTag® ligands) for all determinations of energy transfer activity by this material and its derivatives; or
- 5.3. Contact Promega to obtain a license for the use of the material and its derivatives for energy transfer assays to energy acceptors not manufactured by Promega. No license is needed if the energy transfer acceptor is a genetically encoded autofluorescent protein.

6. For uses of HaloTag® Technology in this material, recipient must either:
- 6.1. Use Promega HaloTag® ligands, which can be modified or linked to Promega or customer-supplied moieties; or
- 6.2. Contact Promega to obtain a license if Promega HaloTag® ligands are not to be used.
