Measure the Ability of Antibodies to Neutralize Viral Entry

Neutralizing antibodies, whether generated through vaccination or produced as therapeutic monoclonal antibodies, are critical for stopping acute and chronic viral infections. Filoviruses like Ebola and Marburg are important targets for researchers developing vaccines, prophylactic monoclonal antibodies or small-molecule inhibitors. Researchers need to determine the neutralizing capacity of their therapeutic, but current methods like PRNT or PNA require infection-competent virus, stringent biosafety measures and cannot be readily implemented in most laboratories.

There are several HiBiT-PsVLP Bioassays designed for Filoviruses that utilize NPC1 for host cell entry. They provide a simple, easily implemented and robust system that can be used to measure the viral neutralization activity of both antibody and small-molecule inhibitors that block viral entry into target cells. The assay consists of a PsVLP from the Filovirus family, such as Ebola or Marburg virus, and a target cell line expressing NPC1. The PsVLP and Target Cell each contain one piece of a split luciferase technology to provide a low-background and convenient luminescent readout.

What is HiBiT?

Principle of Filovirus HiBiT-PsVLP Bioassays. 1. HiBiT-tagged VLPs pseudotyped with the virus-specific surface glycoprotein are added to the (LgBiT) Target cells. 2. In the presence of inhibitors or neutralizing antibodies (nAbs) of Filovirus entry, the entry/fusion processes of PsVLPs are blocked, thereby preventing HiBiT release, and no luminescent signal is produced. 3. In the absence of inhibitors or nAbs, HiBiT-PsVLPs bind to target cells via NPC1 and undergo membrane fusion. HiBiT is released into target cells and binds to LgBiT to generate a luminescent signal.

Filovirus HiBiT-PsVLP Bioassays are suitable for blocking entry of viral particles using nAbs and small-molecule inhibitors. Virus-specific PsVLPs and HEK293T (LgBiT) Target cells were incubated with either anti-ZEBOV GP antibodies (Panel A), anti-SEBOV GP antibodies (Panel B) or anti-MARV GP antibodies (Panel C). After incubation, the Bio-Glo-NB™ VLP Reagent was added and luminescence determined in a GloMax® Discover Plate Reader.
