Reporter Gene Assays for the Investigation of Signal Transduction at Orexin Receptors

Abstract
The neuropeptides orexin-A and -B are involved in the regulation of the circadian rhythm in mammals. Approximately 20-30% of people in western cultures suffer from insomnia. Therefore, the associated orexin-receptor system (OxR-system) is of strong pharmacological interest in drug discovery. The cell signaling receptors OxR1 und OxR2 belong to the family of G protein-coupled receptors. The luciferase reporter assay is an ideal tool to analyze the activity of OxR-ligands and affected cell signaling cascades.
