Protein Purification from Mammalian Cells

The HaloTag® Mammalian Protein Purification System purifies HaloTag® fusion proteins from mammalian cell culture lysates. HaloTag® fusion proteins form a highly specific covalent bond with HaloLink™ Resin, providing an excellent method for purifying recombinant proteins from cultured mammalian cells, even at low expression levels. The specificity and strength of the HaloTag-ligand bond provide significant improvements in purity and functional protein yield over other common purification methods, such as FLAG® tag or His tagged protein purification.

  • Highly pure protein with minimal effort
  • Superior recovery compared to FLAG® and His tag methods
  • Scalable from µg to mg levels
  • Convenient fluorescent protein quantitation method

Highly Pure Protein with Minimal Effort

Highly Pure Protein with Minimal Effort

Five HaloTag-fused human kinases were transiently transfected, expressed and purified from HEK293T cells (120ml culture)1.

Superior Protein Recovery and Purity

The covalent binding of HaloTag®-fusion proteins enables efficient protein recovery and allows stringent washing, which eliminates most non-specific binding. This allows better recovery of highly purified protein compared to FLAG® and His6 tag methods.

Superior Protein Recovery and Purity

Two human kinases were purified from HEK293T cells with four commonly used fusion tags: HaloTag, FLAG, 3xFLAG and His6 tag1
