We describe here our development of a bioluminescent murine FcgRIV-specific ADCC reporter assay. ADCC pathway activation by target cell-bound antibodies is achieved via FcgRIV-mediated NFAT activation and consequent induction of reporter luciferase activity in dual engineered Jurkat effector cells. We have further developed the assay with thaw-and-use effector cells for immediate use, reducing assay variability and providing convenience.
