Homogeneous Assay Measures Caspase-3/7 Activity

The Caspase-Glo® 3/7 assay provides a proluminescent caspase-3/7 DEVD-aminoluciferin substrate and a proprietary thermostable luciferase in a reagent optimized for caspase-3/7 activity, luciferase activity and cell lysis. Adding the single Caspase-Glo® 3/7 Reagent in an "add-mix-measure" format results in cell lysis, followed by caspase cleavage of the substrate. This liberates free aminoluciferin, which is consumed by the luciferase, generating a "glow-type" luminescent signal that is proportional to caspase-3/7 activity.

The stabilized luciferase and proprietary buffer system improve assay performance across a wide range of assay conditions, and the assay is less likely to be affected by compound interference than fluorescent- or colorimetric-based assays. The caspase activity assay is designed for use with multiwell plate formats using either purified enzyme or cells in culture and can be multiplexed with other cell-based assays.  The assay delivers excellent Z´-factor values in cell and purified enzyme models.

Luminescence Is Linear Over a Broad Range of Cell Numbers

Jurkat cells were treated with anti-Fas mAb for 4.5 hours to induce apoptosis or were left untreated. Caspase-Glo® 3/7 Reagent was added directly to the cells in 96-well plates and incubated for 1 hour before recording luminescence. Each point represents the average of 4 wells. The "no cell" blank control value has been subtracted from each.


Multiplex Caspase 3/7 with Cytotoxicity and Cell Viability Assays

Jurkat cells (10,000 cells/well) were treated with serially diluted staurosporine. Cell viability, cytotoxicity and apoptosis were assayed after 6 hours using the ApoTox-Glo™ Triplex Assay, featuring the Caspase-Glo® 3/7 Assay chemistry. Fold change from the untreated cells control (concentration = 0M) was calculated for all data points.

Using 100μl of Caspase-Glo® Reagent per assay in a 96-well format, Cat.# G8090 provides sufficient reagents to perform 25 caspase activity assays; Cat.# G8091, 100 assays; Cat.# G8092 and G8093, 1,000 assays. Using 25μl of Caspase-Glo® Reagent per assay in a 384-well format, Cat.# G8090 provides sufficient reagents to perform 100 caspase activity assays; Cat.# G8091, 400 assays; Cat.# G8092 and G8093, 4,000 assays.
