HaloTag® Ligands
The Promega HaloTag® Ligand portfolio enables a range of functionalities, including affinity labeling for protein purification. These ligands form highly specific and nearly irreversible covalent bonds with HaloTag® fusion proteins under physiological conditions, ensuring stable and efficient labeling with minimal background interference. HaloTag® Ligands enable you to study any compound or surface with a compatible chemical group. With a full suite of HaloTag® Ligand Building Blocks, the possible applications are endless.
HaloTag® Biotin Ligands enable efficient biotin labeling for protein capture and analysis. The HaloTag® Biotin Ligand is cell-permeant, allowing intracellular labeling while unbound ligand can be washed away after incubation.
For enhanced pull-down performance, the HaloTag® PEG-Biotin Ligand offers superior interaction with streptavidin due to its extended and flexible spacer. While it requires cell lysis prior to labeling due to limited membrane permeability, this extended linker can help preserve the activity of HaloTag® fusion proteins during immobilization or derivatization.

Gel Analysis of HaloTag-Based Fusion Proteins

Efficient capture of a p65-HaloTag® fusion protein and co-capture of IκB on paramagnetic streptavidin-coated particles. CHO-K1 cells transiently transfected with plasmid encoding the p65-HaloTag® fusion protein were incubated in the presence or absence of 25µM HaloTag® Biotin Ligand. Cells were lysed, and biotin-labeled proteins were captured on Streptavidin-coated MagneSphere® Paramagnetic Particles (Cat.# Z5481). Proteins were resolved by SDS-PAGE and analyzed by Western blot with anti-p65 antibody (BD Bioscience, Panel A) or anti-IκB antibody (BD Bioscience, Panel B). Content of lanes for both Panels A and B: C = cell lysate; UB = unbound protein; B = bound protein.
