Serial dilution of eGFP mRNA results in uniform decreases in signal intensity. AsPC-1 (left), Jurkat (middle) and HCT-116 (right) cells were transfected with either a 4-fold dilution series of eGFP mRNA (top) or DNA plasmid (bottom). Transfection efficiency and GFP signal intensity were measured by flow cytometry. The tighter peaks after mRNA transfection indicate the transfected cells express at similar levels, and the rightward shift in the peaks with dilution indicate proportionate changes in expression levels across the cell population. Neither of these features were observed with DNA plasmid transfection. The percent GFP positive cells for each condition is listed in the tables.
Efficient, Low-Toxicity Transfection Across Diverse Cell Lines
ViaScript™ mRNA Transfection Reagent achieves high mRNA transfection efficiency with minimal cell toxicity, outperforming DNA transfection in multiple cell lines. A549, CHO, DLD-1, H1299, U2-OS, A375 and Jurkat cells were transfected with mRNA or DNA plasmids. Transfection efficiency was assessed by luminescence, showing strong expression with mRNA transfection. Cell viability remained high across most cell lines, demonstrating the reagent’s low toxicity.
