Why Maxwell® HT?

Maxwell® HT chemistries allow automated extraction of nucleic acids from a range of sample types. Based on the successful chemistry contained in Maxwell® prefilled reagent cartridges, Maxwell® HT Systems allow purification of DNA or RNA at scale on laboratory liquid handlers in 24- or 96-well SLAS format. The easy-to-use method produces the highest quality DNA and RNA, and ensures consistent, reliable results in downstream assays.

Maxwell® purification chemistries use novel magnetic particle-based solutions that naturally decrease contamination carryover.

High-throughput purification of high-quality, amplifiable ccfDNA

The Maxwell® HT Circulating Cell-Free DNA Kit is designed for optimal high-throughput, automated purification of circulating cell-free DNA (ccfDNA) from up to 96 samples of plasma with input volumes of 1–8mls. Protocols are available for the Hamilton® Microlab STAR and Thermo Scientific™ KingFisher™ Flex.

Here, circulating cell-free DNA was purified from 1–8ml plasma samples using the Maxwell HT ccfDNA custom chemistry automated on a Hamilton STAR® and the Qiagen QIAmp® Circulating Nucleic Acid Kit; Qiagen cannot process 8ml. ccfDNA was quantitated by qPCR. Dye-based fluorescence can overestimate the amount of small DNA present due to the presence of large DNA or RNA carrier molecules.

The Maxwell® HT Viral Total Nucleic Acid Kit is designed for high-throughput automated extraction of viral total nucleic acid (both DNA and RNA) from serum, plasma and other samples. Protocols are available for Beckman Biomek® 4000/FX/NX, Eppendorf epMotion® Series, Tecan Freedom EVO®, Thermo Scientific™ KingFisher™ Flex and Hamilton® Microlab STAR.

5,000 IU/ml of inactivated HIV-1 or 500 IU/ml of inactivated hepatitis B virus (HBV) were isolated from 300 μl sample of human plasma using the Maxwell® HT Viral TNA chemistry custom automated on a Tecan Freedom EVO®. Quantitation was performed with PathID® RT-qPCR (HIV-1) and Plexor® RT-qPCR (HBV) using a single-stranded oligonucleotide standard. These proof of concept data were presented as a scientific poster at the 27th Clinical Virology Symposium in 2011.

The Maxwell® HT simplyRNA kit is designed for RNA purification from a variety of sample types to include, but not limited to blood, cells and tissue (fresh/frozen) by high throughput, robotic liquid handler. Protocols are available for Beckman Biomek® 4000/FX/NX, Eppendorf epMotion® Series, Tecan Freedom EVO®, Thermo Scientific™ KingFisher™ Flex and Hamilton® Microlab STAR.

Following processing on the Biomek® 4000 Laboratory Automation Workstation with the magnetic particle-based purification technology, RNA from each tissue type (N=3 or 4) was analyzed using the NanoDrop 1000 spectrophotometer. >8ug of RNA was recovered from kidney, liver, and spleen tissue. The RNA was of high purity as denoted by the A260/A280 and A260/A230 purity ratios. Exceptions were A260/A230 ratios for brain (lipid-rich) and heart (fibrous) tissue, although this slight decrease in purity had no effect in RT-qPCR analysis.
