PKA Kinase Enzyme System

See All Kinase Enzyme Systems
Recombinant bovine cAMP-Dependent Protein Kinase (PKA), Catalytic Subunit, is expressed and purified from E. coli. Intracellular targets include ion channels (1), transcriptional activator proteins (2) and regulatory enzymes of glycogen metabolism (3). PKA does not require cAMP for activity, and the gene sequence can be found at accession number NM_174584.

ADP-Glo™ Kinase Assay is a luminescent kinase assay that measures ADP formed from a kinase reaction; ADP is converted into ATP, which is a substrate in a reaction catalyzed by Ultra-Glo™ Luciferase that produces light. The...

Storage Buffer

350mM potassium phosphate (pH 6.8), 0.1mM DTT.

Storage Conditions

Upon receipt, centrifuge the kinase and dispense it into smaller quantities. Store the kinase at –70°C and the rest of the components at –20°C.

Unit Definition

One unit is the amount of enzyme required to incorporate 1pmol of phosphate into casein in 1 minute. The assay buffer is 40mM Tris-HCl (pH 7.4), 20mM magnesium acetate, 0.2mM ATP and 30,000cpm/μl [γ-[32P] ATP].
