Analyze Challenging Samples and Confirm Your Results

The PowerPlex® ESX 17 and ESI Pro Systems were developed for human identification applications including forensic analysis, relationship testing and research use and meet the new ENFSI recommendations for DNA profile sharing across Europe. They are based on five-color technology and allow co-amplification and detection of 17 loci, including D18S51, D21S11, TH01, D3S1358, Amelogenin, D16S539, D2S1338, D1S1656, D10S1248, FGA, D8S1179, vWA, D22S1045, D19S433, D12S391, D2S441 and SE33.

These two STR systems are available separately or as a bundle and can be used to complement each other when analyzing degraded or challenging samples to maximize recovery of allelic information from as many loci as possible and also allow confirmation of results obtained with the other system.

These systems include a proprietary 5X master mix that contains hot-start Taq DNA polymerase, buffer, salts, MgCl2 and dNTPs. Additionally, the kits have increased tolerance to common inhibitors and increased sensitivity to obtain full profiles from low-level DNA and are robust enough to genotype degraded DNA samples through the use of mini-STR loci.

The PowerPlex® ESX 17 and ESI 17 Pro Systems are compatible with ABI PRISM® 310, 3100 and 3100-Avant Genetic Analyzers and Applied Biosystems® 3130, 3130xl, 3500 and 3500xL Genetic Analyzers. Panels and bins text files are required for use with GeneMapper® ID and ID-X software and are available for download.

PowerPlex® ESX 17 System

The PowerPlex® ESX 17 System amplifies the loci recommended by the European Network of Forensic Science Institutes (ENFSI) and European DNA Profiling Group (EDNAP) as mini-STRs (<125bp; D2S441, D10S1248 and D22S1045) or midi-STRs (125–185bp; D1S1656 and D12S391).

A DNA sample (500pg) heterozygous at all loci was amplified in the presence of 300µM hematin in a 25µl reaction with the PowerPlex® ESX 17 System for 30 cycles using a GeneAmp® PCR System 9700 thermal cycler. A loading cocktail containing CC5 Internal Lane Standard 500 and formamide was mixed with 1µl of amplified sample, and amplified fragments were detected using an Applied Biosystems 3130 Genetic Analyzer and a 3kV, 5-second injection. Data were analyzed using GeneMapper® ID software, version 3.2.1, and PowerPlex® ESX panel and bin files.

PowerPlex® ESI 17 Pro System

The PowerPlex® ESI 17 Pro System is designed to amplify six of the original seven European Standard Set (ESS) loci (D3S1358, D18S51, TH01, vWA, D8S1179 and the more common FGA alleles) along with D16S539 and D19S433 as smaller amplicons (<250bp), while the loci recommended by the European Network of Forensic Science Institutes (ENFSI) and European DNA Profiling Group (EDNAP) (D1S1656, D2S441, D10S1248, D12S391 and D22S1045) are present as larger amplicons.

The PowerPlex® ESI 17 Pro System is an update to the PowerPlex® ESI 17 System. All primer pairs, with the exception of the SE33 primer pairs, are identical in both systems. The internal lane standard, WEN Internal Lane Standard 500 ESS, is designed and optimized specifically for this system.

The 5X AmpSolution™ Reagent, SwabSolution™ Kit and PunchSolution™ Kit allow fast and simple processing of swabs and punches for analysis with various PowerPlex® Systems, including the PowerPlex® Fusion, 21, 18D, 16 HS, ESX, ESI, CS7 and Y23 Systems. These products are intended for preparation of single-source reference, database and paternity samples where sample purification is unnecessary.
