Comparision of Promega’s M-MLV RT, RNase H Minus, Point Mutant
(P) and SuperScript™ II (S) for synthesis of full length cDNA
from a 7.5kb RNA. First-strand cDNA reactions were performed
using a 7.5kb RNA template (1µg in 1µl), 0.5µg of oligo(dT)15
and 15.25µl of water in a 0.5ml microcentrifuge tube heated at
70˚C for 5 minutes. The reactions were then cooled on ice for 5
minutes before adding 5µl of M-MLV RT 5X Buffer, 1.25µl of 10mM
dNTPs, 0.5µl [α-32P dCTP] (10µCi/µl, 400Ci/mmol), and 200 units
of reverse transcriptase. The reactions were incubated at 40˚C
for 50 minutes. Samples were then separated on an alkaline
agarose gel and autoradiography was performed. |
