About the P450 -Glo System
This systems provide a complete set
of reagents for performing luminescent P450 assays. Systems are
available for
CYP1A2 ,
CYP2C9,
CYP3A4,
CYP3A4 and
CYP2D6. These assays
are ideal for testing the effects of drugs and new chemical entities
on
native and recombinant P450 activities.
Conversion of the P450-Glo™ substrate to luciferin by cytochrome P450
The P450-Glo™ assays employ luminogenic cytochrome P450 substrates that
are derivatives of beetle luciferin, a substrate for firefly luciferase.
Cytochrome P450s convert the substrates to luciferin, which is acted on
by luciferase to produce light that is directly proportional to the
activity of cytochrome P450.

Superior Sensitivity
The P450-Glo™ Screening Systems have low background levels and
large dynamic ranges. Less cytochrome P450 is required in these
assays than in typical HPLC or fluorometric methods because of
the enhanced sensitivity.

Accurate IC50
determinations using P450-Glo™ assays
In addition to detecting inhibitors of cytochrome P450
enzymes, a P450 assay must be able to assess inhibitor potency
by providing accurate IC50 values.
Inhibitor titrations were
performed with CYP2C9 and Luciferin-H in the presence of
increasing concentrations of diclofenac (Panel A) and with
CYP3A4 and Luciferin-BE in the rpesence of increasing
concentration of ketoconazide (Panel B)
