Immunocytochemistry of Human Fetal Microglia Cells Using the Anti-BDNF pAb

Culture microglia cells on coverslips
Remove media and gently wash cells 2 times with PBS
Fix cells in 4% paraformaldehyde at room temperature for 30 minutes
Wash cells 2 times with PBS.
Apply Promega's Anti-Human BDNF pAb diluted 1:100 in Common Antibody Diluent (CAD; Biogenex Laboratories)
Incubate at room temperature on an orbital shaker for 2 hours
Wash cells 3 times with PBS
Apply rabbit anti-chicken IgY-DTAF conjugate (Jackson ImmunoResearch Laboratories) diluted 1:100 in CAD.
Protect from light and incubate at room temperature for 30 minutes
Rinse cells once with PBS and mount on glass slides with Gelvatol fluorescent mounting medium.
