Anti-Human Tryptase mAb–Biotin Conjugate

Tryptase, a 135kDa tetrameric serine protease, is the principal protein mediator component of mast cell granules and is abundant in both degranulated and unstimulated mast cells. It therefore is an exquisite marker in heterogeneous populations of mast cells. The Anti-Human Tryptase mAb Biotin (Cat.# G3361) allows for positive phenotypic identification of mast cells in disease of many organs and has been qualified in Western blot analysis, ELISA and immunocytochemistry applications with complex biological fluids. This antibody is both more specific and sensitive than standard metachromic staining procedures for mast cells. Furthermore, it is effective for a wide range of both Carnoy's and formaldehyde-fixed tissues. The biotinylated tryptase mAb allows for amplification of signal with a streptavidin conjugate (e.g., HRP or FITC) while maintaining very low background from nonspecific reactivity, a problem often associated with use of secondary antibodies.

  • Antibody Form: IgG1 (clone AA5), affinity purified; supplied as 1mg/ml in PBS containing no preservatives.
  • Specificity: No cross-reactivity to related biomolecules; cross-reacts in non-human primates.

ELISA

The antibody has been used in an indirect ELISA to recognize rhTryptase.

The antibody was used with another antibody in a sandwich ELISA to quantitate tryptase by Okragly, A.J. et al. (1999) Elevated tryptase, nerve growth factor, neurotrophin-3 and glial cell line-derived neurotrophic factor levels in the urine of interstitial cystitis and bladder cancer patients. J. Urol. 161, 438–441. The other antibody described in the paper, an IgY, is not commercially available from Promega or any company.
