Abstract for PowerPlex® 2.1 System
The PowerPlex® 2.1 System allows the co-amplification and
two-color detection of nine STR loci. The PowerPlex® 2.1 System
contains the loci Penta E, D18S51, D21S11, TH01, D3S1358, FGA, TPOX, D8S1179 and
vWA. One of the two primers for Penta E, D18S51, D21S11, TH01 and D3S1358 is
labeled with fluorescein (FL) and one primer specific for FGA, TPOX, D8S1179 and
vWA is labeled with carboxy-tetramethylrhodamine (TMR). All nine loci are
amplified simultaneously in a single tube and analyzed in a single gel lane.
The PowerPlex® 2.1 System is customized for use with the Hitachi
FMBIO® and FMBIO® II Fluorescent Scanners. The system
provides all of the materials necessary for amplification of STR regions of
purified genomic DNA with the exception of AmpliTaq Gold® DNA
polymerase. This manual contains separate protocols for use of the PowerPlex®
2.1 System with the Perkin-Elmer model 480 and GeneAmp® PCR system
9600, 9700 and 2400 thermal cyclers, in addition to protocols for separation of
amplified products and detection of separated material. Protocols for operation
of the fluorescence-detecting instrumentation should be obtained from the
manufacturer.
