Abstract for A Luminescent Cytotoxicity Assay for Drug Discovery

We have developed a non-lytic, homogeneous, luminescent assay
method number of features useful for cytotoxicity testing in high
that embodies a number of features useful for cytotoxicity testing
in high density multi-well plate formats. This assay measures a
ubiquitous and conserved proteolytic activity corresponding with
loss of cellular membrane integrity. The assay demonstrates superior
performance with respect to sensitivity and useful linear range of
cell numbers when compared with other commercially available
luminescent cytotoxicity assays. The assay chemistry also
demonstrates an improvement in marker half-life and resulting
luminescent signal stability. Lastly, the assay can be conveniently
coupled with a fluorescent measure of viability to multiplex
viability and cytotoxicity assays which offers additional utility
for response normalization and determination of compound specific
mode-of-action.
