The ability to provide simple solutions for correlating multiple parameters within cellular processes is essential for today’s screening facilities. With a variety of plate readers available to laboratories, it is necessary to validate assays in a variety of testing situations and plate formats. Here we demonstrate the scalability of the Chroma-Luc™ Technology, a dual color luminescent reporter methodology that generates red and green luminescence upon addition of a single reagent.
Cellular lysates, containing the green CBG99luc and red CBRluc genes, and reagent were dispensed manually in 96 and 384-well formats, or using a noncontact dispenser for1536-well format. Data was collected with Molecular Devices and PerkinElmer® HTS plate readers. The excellent linearity, limit of detection, separation of signal, and Z’ Factor scores show the ability of this chemistry to generate a stable dual color signal and be detected on a variety of HTS plate readers used in high-throughput screening environments.
