Introduction to the READIT® SNP Genotyping System: Reaction Mechanism

Let’s begin by reviewing the enzymatic activities of DNA polymerase, as it is central to the READIT® Technology. This slide is an overview of the forward, or polymerization reaction carried out by DNA polymerase. In this reaction, polymerase catalyzes a 3´ hydroxyl attack on the alpha phosphate of the incoming deoxynucleoside triphosphate. The net reaction given at the bottom of the slide is extension of the 3´ end of the DNA template in the presence of deoxynucleoside triphosphates, with the release of inorganic pyrophosphate.
