Getting Started with the READIT® System
Genotyping begins with sample processing and purification of nucleic acids. Amplification by PCR or RT-PCR gives a product ready for the READIT® Assay. The initial step of the READIT® Assay is PCR product purification. Purification usually means lengthy column or resin-based processes. Promega has developed
a new enzymatic method to prepare the PCR product for molecular interrogation
replacing traditional purification methods. Purified PCR products are added to the READIT® Interrogation reaction resulting in bioluminescent signals collected by a luminometer.
Data is sent to the READIT® Calculator resulting in a genotype call.
The READIT® Assay involves a simple set of reagent additions and transfers.

You have the flexibility of choosing manual evaluation requiring a multi-channel pipette, a heating block and a plate-reading luminometer or an automated evaluation involving a robotic liquid handler, heating block and luminometer. Promega provides the READIT® System Verification Module (Cat.#MD1220) containing a practice target template and interrogation probes to use to familiarize yourself with the workflow of the Assay. Once you have experienced the minimal pipeting, robustness of the chemistry and ease of data processing, you can move to designing your own probes for genotyping your own samples.

First contact us to discuss your options. You can chose to attend a training course to quickly gain the knowledge about the READIT® Assay. Completion of training leaves you with the confidence you can begin genotyping your samples when you return home. You can choose to work with the System Verification Module to quickly begin working with the Assay in your own lab. Technical Service assistance is available by phone, e-mail or fax to help get you going. Finally, you may request Promega's Applications Scientists develop and assay for you. Everyone is different and the READIT® System offers you the versatility of creating your own assay. If you need help, the on-line probe design software and our technical services scientists can help. You choose how you would like to implement the READIT® technology in your lab.
