Know
your results are right!
Not all genotyping methods are equal when it comes to accuracy and
reliability. The READIT® System is inherently accurate,
combining three independent approaches to deliver a genotype result:
hybridization at elevated temperature, thermostable enzyme fidelity
and integrated data calculation with error checking.
When the 3´-end of the interrogation probe is perfectly hybridized
to one strand of a PCR product, it becomes a substrate for the
READIT® reaction. The READase™ Polymerase, an
engineered thermostable DNA polymerase, releases dNTPs from the
3´-end of the hybridized probe. Signal generation occurs through a
series of coupled enzyme reactions.

No light signal will be generated if there is a mismatch between the
interrogation probe and the PCR product, allowing accurate and
reliable genotyping results. Use of thermostable enzymes with high
fidelity and protocols specifically optimized to minimize
variability lead to an assay with unprecedented accuracy.

Statistical confidence in the results and unambiguous determination of the
genotype allows development of assays with a high degree of reliability.
Typical six sigma separation between genotypes provides confidence that
genotype calls will be correct.

A retrospective analysis of over 1,200 samples for the Factor V Leiden genotype
showed 100% concordance with PCR/RFLP
genotyping methods.

Data
analysis using the READIT® Calculator
Automation of data analysis eliminates subjective genotype calls.
The READIT® Calculator checks each analysis for known
sources of error before determination of the genotype. Instead of
designating an incorrect genotype, the calculator flags inconsistent
samples for further analysis. You choose whether to print your
results, convert them to an electronic file or study them further
with the statistical analysis functions of the READIT®
Calculator.
