HaloTag® Custom Vectors and Ligands

HaloTag® technology is a comprehensive platform for functional protein analysis based on the formation of a covalent bond between a protein fusion tag (HaloTag) and synthetic chemical ligands. By using different ligands you can control the function and properties of the HaloTag® fusion protein. The HaloTag® system gives exceptional performance in cell-based and biochemical assays without the need to switch fusion tags or reclone inserts.

In addition to the HaloTag® Vectors and Ligands available as catalog items, we offer a selection of pre-built custom HaloTag® vectors and ligands developed by our Custom Assay Services team within R&D. Although functionally tested, they are not manufactured under the Promega ISO13485 quality system and are sold “as is” with no warranty either stated or implied.

HaloTag® Vectors

N-terminal and C-terminal HaloTag® Flexi® Vectors for attenuated expression, protein secretion and nuclear localization in mammalian cells, and vectors containing regulated mammalian, TK, SV40, PGK, EF1α promoters are available.

HaloTag® Ligands

  • pH sensor ligand to detect internalization of membrane proteins
  • Eosin ligand for chromophore-assisted laser inactivation (CALI) for targeted protein destruction
  • HaloTag® PROTAC3 (proteolysis targeting chimera) is a permeable small-molecule compound that can target HaloTag® fusion proteins for degradation. HaloTag® PROTAC will bind covalently to HaloTag and induce interactions with E3 ligase machinery proteins, resulting in ubiquitination of the HaloTag® fusion and subsequent trafficking to the proteasome for degradation.
