pGL4 Series Vectors and GloResponse™ Cell Lines

Easily quantify changes in signaling pathways with luciferase reporter assays

The new pGL4 series vectors and GloResponse™ cell lines offer ideal platforms for monitoring entire cell signaling pathways such as NF-kB. Now the modulation of pathways can be studied faster and with more sensitivity than ever before. By providing optimized vectors, cell lines, engineered luciferase genes, and add-mix-read assay reagents, Promega simplifies cell signaling.

Improved Sensitivity and Biological Relevance Due to:

  • Increased Reporter Gene Expression: Codon optimization of synthetic luciferase genes for mammalian expression.
  • Reduced Background and Risk of Expression Artifacts: Removal of cryptic DNA regulatory elements and transcription factor binding sites.
  • Quick Response: Rapid Response™ technology using destabilized luciferase genes.

Figure. Automated ONE-Glo™ Assay dose-response curve. Dose-response curves were generated using TNFα-treated (Panel A) and anti-TNFα-treated (Panel B) GloResponse™ NF-κB-RE-luc2P HEK293 cells in 96-well plate format. EC50 value obtained for TNFα-treatment assay was 1.1 ng/ml. ND50 value obtained for TNFα neutralization assay was 78 ng/ml. Z'-factor values for both assays were >0.71.
