The ILS included with the 5-dye PowerPlex® Systems has changed. What are the changes?

The appropriate use of an ILS during capillary electrophoresis allows automated data analysis and correct DNA fragment size assessments between runs. As part of our effort to provide products that exceed your performance expectations, we have included an ILS with WEN, a dye which is brighter and more photostable and can be stored at 4°C, in our PowerPlex® Fusion 6C System. Recently, we have changed the dye used in the ILS in our PowerPlex® 5-dye STR systems as well. The new ILS has the same fragment sizes as the current CC5 ILS 500 fragments, but the fragments are instead labeled with WEN dye.


How does the ILS change affect analysis of PowerPlex® 5-dye STR data?

Fragments labeled with CC5 and WEN migrate slightly differently during capillary electrophoresis, and thus this dye change will require the use of updated panels and bins for correct allele assignment in unknown samples. Also, the slight migration differences will result in changes in predicted sizes of artifacts seen with various 5-dye PowerPlex® Systems.

The figure below demonstrates that although the fragments migrate differently, the allele calls are concordant and correct.

Allelic ladder samples were mixed with either 1–2µl of CC5 ILS500 or 0.5µl of WEN ILS 500 and then injected on an Applied Biosystems® 3500xL Genetic Analyzer with a 1.2kV, 24-second injection. Ladders were analyzed with a 175RFU analysis threshold, and data were analyzed with GeneMapper® ID-X software using the current or updated panels and bins text files.

Does this mean the matrix standards have also changed to include the WEN dye?

Yes, due to the ILS dye change, a corresponding dye change has been implemented in our matrix kits. The matrix standards for multicapillary instruments are now provided in a single-tube format with a recommended storage temperature of 4°C—making it more convenient for users.


What changed in the PowerPlex® STR kits?

The pre-amplification components of the STR system (primer mix and master mix) have not changed. The catalog number and product names of the STR kits also have not changed. The only change is that beginning June 10, 2016, the 5-dye PowerPlex® Systems will contain an ILS that is labeled with WEN dye instead of CC5 dye.
