Promega announces its new Caspase-Glo™ 8 and Caspase-Glo 9 Assays
that deliver unprecedented speed and sensitivity for apoptosis detection.
It is the only homogeneous assay protocol for Caspase 8 & 9 and delivers
results in as little as 20 minutes. The sensitivity is orders of
magnitude greater than fluorescent methods, and these assays enable
laboratories to precisely monitor toxic effects of compounds at
high throughput capacity or one sample at a time.
The Caspase-Glo Assay protocol consists of a single-step-reagent
addition of a lysis/activity buffer containing a peptide-aminoluciferin
caspase substrate and a thermostable luciferase. In the presence
of caspase, the peptide is cleaved releasing aminoluciferin for
use by luciferase, resulting in a “glow-type” luminescent signal
that is proportional to caspase activity.
The buffer allows rapid and efficient cell lysis while supporting
optimal caspase and luciferase activities and eliminates sample
extract preparation steps. The bioluminescent assays avoid potential
problems associated with auto fluorescence and fluorescent interference
encountered in fluorescent-based cell assays and compound library
screens. The low background provides excellent signal to background
ratios and exquisite sensitivity. The superior sensitivity allows
scientists scale assays to 96 or 384-well plates and cut costs because
less compound and fewer cells or less purified caspase is required.
A stable luminescent signal allows batch processing of plates in
multiwell format; no reagent injectors required.
