The PowerPlex S5 System is an extremely sensitive STR kit that allows you to screen forensic casework quickly and cost-effectively. Reproducible STR results can be obtained from as little as 50pg of DNA (see Figure 1 below). The system includes 4 STR loci: D18S51, D8S1179, TH01, and FGA, plus the gender-determination marker Amelogenin. All amplified fragments are less than 260bp, including the extended alleles from FGA. Allelic ladder is included in the kit.

One microliter of the amplification was analyzed on an Applied
Biosystems 3130xl Genetic Analyzer with a 3kV, 5-second injection.

PowerPlex S5 is the first STR system from Promega to include Taq polymerase, and like all other Promega STR systems, PCR setup takes place at room temperature. The PowerPlex S5 System is compatible with the ABI PRISM® 310, 3100 and 3100-Avant Genetic Analyzers, and Applied Biosystems 3130 and 3130xl Genetic Analyzers. For PCR amplification, the system is optimized for use with the Applied Biosystems GeneAmp® PCR System 9700 and is compatible with the 2720 Thermal Cycler. Internal lane standard is included. For a full description of the protocol and system components, please refer to the PowerPlex S5 System Technical Manual.

In each panel, 250pg of template DNA was amplified in the presence
of the indicated concentrations of hematin.

The PowerPlex S5 System is extremely resistant to inhibitors such as heme, humic acid, and dyes (see Figure 2 above). The mini-STR format of the system includes significantly reduced allele sizes for the TH01, D18S51, and FGA loci (see Figure 3 below), allowing you to gain more information from degraded or inhibited samples.
