Isolation and Analysis of Nucleic Acids from Human Blood: A Molecular Diagnostics Application

By Daniel Kephart, Ph.D., and Hemanth Shenoi, Ph.D.
Promega Corporation

Rapid processing of blood samples is critical for maximizing the yield and quality of isolated nucleic acids. However, the appropriate processing of blood samples may be impractical or impossible with samples such as archived blood. This report demonstrates the successful isolation of total RNA from frozen whole blood using Promega's SV Total RNA Isolation System. However, when compared to blood treated to remove nonleukocyte material prior to freezing, the unprocessed, frozen whole blood yields less RNA and RNA that does not perform as well in RT-PCR .
