Assay Systems for Detecting Apoptosis and Cell Death

By Rich Moravec and Terry Riss
Promega Corporation

Most eukaryotic cells have the ability to self-destruct by activation of an intrinsic cellular suicide program referred to as programmed cell death or apoptosis. Many assay systems are available for detecting apoptosis, providing the ability to measure various parameters associated with the cell death process. This study illustrates the utility of the CellTiter 96® AQueous One Solution Cell Proliferation Assay, the CytoTox 96® Non-Radioactive Cytotoxicity Assay, the CaspACETM Assay System, Fluorometric, and the Apoptosis Detection System, Fluorescein, as methods to elucidate apoptotic events. These four systems measure viable cell number, cell membrane damage, caspase activation and DNA fragmentation, respectively.
