Increased Expression and Convenience with the New pGL3 Luciferase Reporter Vectors

The pGL3 Vectors are an improved family of reporter vectors utilizing the firefly
luciferase gene and based on the previously developed pGL2 Reporter Vectors. The pGL3
Vectors contain the new cytosolic form of luciferase (luc+) and multiple changes to
the vector backbone to support efficient reporter expression and greater user convenience.
These new vectors yield 10- to over 100-fold greater expression of luciferase in mammalian
cells, depending on the cell line. Here we describe the rationale underlying the new
vector design and present data comparing the pGL3 to the pGL2 Vectors.
