Development of Antibodies Reactive with the gamma-c (Common gamma) Chain of the Human
Hematopoietic Receptor Superfamily

We tested two preparations of a polyclonal antibody developed in chicken to react
with the human gamma-c chain. We performed immunoprecipitations on cell lysates from the
YT leukemic line which expresses the IL-2 Receptor beta-gamma-c complex and compared
results using the two preparations of polyclonal chicken antibodies to those generated
using the mouse monoclonal antibody 561 (mAb 561) which recognizes the beta chain of the
IL-2 Receptor complex and can co-precipitate gamma-c when the IL-2 Receptor has bound
IL-2. The chicken polyclonal antibody precipitated a surface molecule that corresponded to
the gamma-c spot (by 2D gel analysis) based on results with the mAb 561, demonstrating its
effectiveness as a reagent for studying gamma-c.
