Amber suppressor tRNAs have been isolated which allow insertion of any one of 12
amino acids at an amber (TAG) stop codon. Using a set of strains containing these various
suppressor tRNAs, it is possible to determine the effect of substituting each of these 12
amino acids at a particular site by making a single amber mutation.
