RNase ONE offers several advantages over other enzymes used in nuclease protection assays. It cleaves 3´ to all bases, enabling more accurate mapping than with mixtures of RNase A and T1. Because of the high purity of cloned RNase ONE, less optimization is required to obtain a reproducible signal level. RNase ONE also is inhibited by 0.1% SDS, permitting ethanol precipitation of the protected RNA fragments without prior phenol:chloroform extractions.
